TWIST Bioscience Targeted sequencing service
CNRGV provides a specialized service for targeted sequencing of genomic regions of interest, combining sequence capture with long-read sequencing technologies. This service is tailored to characterize regions of interest (ROI) ranging from ten to several hundred kilobases across multiple varieties, constituting a diversity panel.
The approach leverages capture technology using probes from TWIST Bioscience company, specifically the "Twist Custom Target Enrichment Panels," along with long-read sequencing platforms, PacBio or ONT. This combination enables the high-quality assembly of ROIs spanning tens of kilobases, providing detailed insights into structural variations among the varieties of the diversity panel.
CNRGV manages the entire workflow, from designing the probe panel in collaboration with TWIST Bioscience, to DNA extraction, sample preparation, sequencing, and the assembly of sequence data.
To implement this method, a reference genome and the sequence of the ROI are required. This reference serves as the basis for designing probes that cover the entire locus. Using this sequence, the CNRGV team will evaluate your project and deliver a feasibility assessment based on the ROI’s characteristics, especially its ratio of specific to repeated sequences.
